Technical requirements for cleanliness testing laboratory
The cleanliness laboratory mainly uses manual methods to control the dust, bacterial concentration, temperature, humidity and pressure in the indoor air to achieve the required environmental parameters such as cleanliness, temperature and humidity, and airflow speed. Air cleanliness refers to the degree of dust in a clean air environment, which is divided by the dust concentration. Cleanliness refers to the total particles of dust particles with a particle size of ≥0.5 microns per liter of air. China's air cleanliness standards are divided into: 100, 1000, 10,000, and 100,000.
International standards are divided into: 1, 2, 3, 4, 5. Cleanrooms typically achieve two levels of isolation. Primary isolation is achieved through biological safety cabinets, negative pressure isolators, positive pressure protective clothing, gloves, goggles, etc.; secondary isolation is achieved through laboratory construction, air conditioning purification and electrical control systems.
However, since the air volume required for air conditioning purification is small, the air volume is estimated to be large, generally exceeding the specifications, and the performance is more than 100. The cooling and heating load may be estimated under the condition that the requirements are not very strict. The unit can be used directly. Two-level isolation should be implemented in the biosafety laboratory of level 2 to level 4. The general laboratory equipment includes: ultra-clean workbench, biosafety cabinet, sideboard or stainless steel console, washing table, etc.
Taking the PRC laboratory as an example, PCR or polymerase chain reaction is a routine method experiment in molecular biology research, and it is also an experimental technique widely used in biology and medical clinical fields, which is characterized by a large increase in trace amounts of DNA. The laboratory is usually divided into four areas, namely: reagent storage and preparation area, sample preparation area, amplification reaction mixture preparation and amplification area, and amplification product analysis area, automatic analyzer, and the areas can be appropriately combined.
Entering each area must be strictly in one direction, and different work areas use different work clothes, such as different colors. Workers should not bring their work clothes when they leave.
